10X ATAC


Service Introduction: 10X Genomics is a microfluidic technology that uses Tn5 transferase to slice open chromatin at a specific time and space and adds millions of unique Barcode labeled single cells to map chromatin accessibility through high-throughput sequencing. The method reveals differences in chromatin accessibility within the nucleus at the single-cell level. It provides data basis for epigenetic analysis at single cell level. 10X ATAC

Cell

The number of cells is 1×107-5×107, and the cell activity is > 90%. It can be sent in a short time. After cell suspension is obtained, the nucleus is extracted and the library is constructed

Blood

Sufficient amount of blood is drawn and added to the anticoagulant tube containing heparin sodium, which can be sent in a short time. After obtaining the blood, the nucleus is prepared immediately and the subsequent library building experiment is carried out (the amount of blood will be adjusted according to the experimental purpose).

Tissue

Take sufficient amount of fresh tissue and send it in a short time. After obtaining the tissue, prepare the nucleus immediately and conduct the subsequent library building experiment (the amount of tissue varies according to the experiment purpose).

Q: What are the requirements for preparing a single cell suspension?


Survival rate: > 90%;


Concentration: 700-1200 cells/μL;


Number: at least 1*107 viable cells (detected with a cell counter for detection);


Diameter: 5-30 μm;


Q: What are the requirements for preparing a nuclear suspension?


Activity: <5%; (try to remove cell debris and cell clumps as much as possible;


Integrity: under the microscope, the nucleus morphologically intact;


Concentration: 700-1200 cells/μL;


Number: at least 20,000 nuclei (detected with a cell counter for detection);


Q: Can the stained nuclei still do 10X ATAC?


No, binding TOD dyes affects the DNA conformation within the nucleus, resulting in biased ATAC results.
